Overview

ALDH1A2 antisense RNA 1 (ALDH1A2-AS1) is a long non-coding RNA (lncRNA) transcribed from the antisense strand of the ALDH1A2 gene locus. As an antisense lncRNA, ALDH1A2-AS1 is presumed to regulate the expression of the neighboring protein-coding gene ALDH1A2, either by direct hybridization with the sense transcript (ALDH1A2 mRNA), by modulating chromatin structure, or through other epigenetic and post-transcriptional mechanisms. Hypermethylation and other epigenetic changes of many antisense lncRNAs, including those similar to ALDH1A2-AS1, have been observed in leukemias and may contribute to dysregulated expression of protein-coding genes, possibly acting as a driving event in malignant transformation or disease progression. Although ALDH1A2-AS1 is not a therapeutic target on its own, its methylation or expression status could be investigated as a biomarker in specific leukemias. The gene "ALDH1A2" encodes an enzyme (ALDH1A2/RALDH2) with important roles in development and cancer; however, "ALDH1A2-AS1" refers to the antisense non-coding RNA and not the enzyme itself. There is no evidence that the antisense RNA is directly druggable or acts as a receptor, enzyme, transporter, etc.

Mechanism of action

No approved or investigational drugs specifically target ALDH1A2-AS1; general lncRNA targeting strategies in preclinical research may include antisense oligonucleotides, RNAi, or epigenetic modulators.

03

Biological functions

Regulation of gene expression (primarily via antisense mechanism)Epigenetic regulation (e.g., DNA methylation affecting transcription)Potential involvement in alternative splicing, chromatin remodeling, miRNA interaction, or recruitment of transcriptional regulators (general to antisense lncRNAs)
04

Disease associations

Cancer (notably B-cell acute lymphoblastic leukemia and possibly other malignancies via regulatory mechanisms)Other (pending specific functional studies, general for lncRNAs in disease)
05

Safety considerations

There are no known or reported safety issues specific to targeting ALDH1A2-AS1, as it is not currently a direct therapeutic target.
06

Biomarkers

Aberrant DNA methylation levels of ALDH1A2-AS1 and related antisense lncRNAs can stratify B-cell acute lymphoblastic leukemia subtypes and may have diagnostic or prognostic value
