Aldo-keto reductase family 1 member B10 (AKR1B10)

Target
AKR1B10
Molecular classification
Enzyme, Oxidoreductase, Aldo-keto reductase superfamily
01

Overview

Aldo-keto reductase family 1 member B10 (AKR1B10) is an NADPH-dependent enzyme of the aldo-keto reductase superfamily that catalyzes the reduction of carbonyl-containing compounds, especially aliphatic and aromatic aldehydes, protecting cells from oxidative stress and regulating lipid metabolism. It is highly expressed in the gastrointestinal tract and overexpressed in multiple cancers, where it has been associated with tumor progression, chemoresistance, and is under investigation as a prognostic biomarker

02

Mechanism of action

Drugs inhibiting AKR1B10 act as enzyme inhibitors, reducing its activity in metabolic detoxification and possibly abrogating chemoresistance in certain cancers

03

Biological functions

Xenobiotic metabolismDetoxification of carbonyl compoundsReduction of aliphatic and aromatic aldehydesLipid metabolismProtection against oxidative stress
04

Disease associations

Cancer (including liver and digestive system cancers)CarcinogenesisChemoresistance
05

Safety considerations

Targeting AKR1B10 could affect detoxification and lipid metabolism, potentially leading to unintended metabolic disturbances or altered drug metabolism; impact on normal tissue expression remains a therapeutic challenge
06

Interacting drugs

No specific approved drugs directly targeting AKR1B10 were identified in the search results. However, AKR1B10 may influence drug resistance or metabolism of chemotherapeutic agents
07

Biomarkers

Overexpression in several cancers (e.g., hepatocellular carcinoma, colorectal cancer) is investigated as a biomarker for cancer prognosis and possibly treatment response
