Overview

Aldo-keto reductase family 1 member B10 (AKR1B10) is a cytosolic NADPH-dependent enzyme that belongs to the aldo-keto reductase superfamily and is primarily involved in the detoxification of reactive carbonyl compounds and the metabolism of retinoids and lipids. Under physiological conditions, it is highly expressed in the gastrointestinal tract, where it protects the mucosal lining from dietary and lipid-derived aldehydes. However, AKR1B10 is significantly overexpressed in several malignancies, including hepatocellular carcinoma, non-small cell lung cancer, and breast cancer, where it promotes tumor cell survival, proliferation, and metastasis. Furthermore, AKR1B10 is a major driver of chemoresistance, as it can metabolize and inactivate various anticancer drugs, such as anthracyclines, and its upregulation is associated with poor clinical outcomes. Due to its restricted expression in normal tissues and high prevalence in tumors, AKR1B10 is an attractive therapeutic target for small-molecule inhibitors and RNA-targeted therapies like siRNA. Additionally, its presence in serum makes it a valuable non-invasive biomarker for early cancer detection and monitoring treatment efficacy.
