Overview

Aldose reductase (AR) is a cytosolic, NADPH-dependent oxidoreductase enzyme that catalyzes the reduction of a wide range of aldehydes and carbonyl compounds, including monosaccharides such as glucose. It is the first and rate-limiting enzyme in the polyol pathway, converting glucose to sorbitol. AR is strongly implicated in diabetic complications due to increased flux through the polyol pathway under hyperglycemic conditions. It is a therapeutic target for preventing or ameliorating these complications.
