Alkaline Phosphatase, Tissue-Nonspecific Isozyme (TNAP)

Target
TNAP
Molecular classification
Enzyme
01

Overview

Alkaline phosphatase, tissue-nonspecific isozyme (TNAP) is an enzyme encoded by the ALPL gene. It catalyzes the dephosphorylation of phosphate esters such as inorganic pyrophosphate (PPi), pyridoxal 5'-phosphate (PLP), and phosphoethanolamine (PEA). It plays a critical role in skeletal mineralization and vitamin B6 metabolism. Mutations cause hypophosphatasia with variable severity, affecting bone and tooth mineralization.

Mechanism of action

Inhibition of TNAP would theoretically prevent dephosphorylation of PPi, PLP and PEA.
