AlkB homologue 3 antisense RNA 1 (ALKBH3-AS1) is a long noncoding antisense RNA located on human chromosome 11p11.2, consisting of approximately 1927 nucleotides. It is expressed in both the nucleus and cytoplasm of hepatocellular carcinoma (HCC) cells, and its upregulation is correlated with poor patient survival. ALKBH3-AS1 exerts its oncogenic effects by stabilizing ALKBH3 mRNA, thereby promoting malignant cellular behaviors such as proliferation and invasion. Its expression is induced under hypoxic conditions via direct transcriptional activation by hypoxia-inducible factor 1-alpha (HIF-1α). Silencing ALKBH3-AS1 leads to reduced ALKBH3 mRNA stability and suppressed tumor growth and invasion in experimental models. ALKBH3-AS1 is considered a potential therapeutic target and biomarker in HCC and possibly other cancers where hypoxia-driven gene regulation is important.
