An allergen-specific T cell receptor (TCR) is a membrane-bound protein complex found on the surface of T lymphocytes (T cells) that specifically recognizes fragments of allergens presented by major histocompatibility complex (MHC) molecules on antigen-presenting cells. These receptors are central to the adaptive immune response in allergic diseases, as they initiate and regulate immune reactions against otherwise harmless environmental antigens known as allergens. The typical TCR is a heterodimer composed of two polypeptide chains: an alpha (α) and a beta (β) chain in about 95% of cases; the remaining 5% consist of gamma (γ) and delta (δ) chains. Each chain has variable regions containing complementary determining regions (CDRs), which provide specificity for antigen binding. Upon allergen recognition, these receptors trigger downstream signaling pathways leading to activation, proliferation, and differentiation of allergen-specific Th2 cells, and ultimately promoting IgE production by B cells.
