Overview

The target refers to the interaction between allogeneic Major Histocompatibility Complex (MHC) class I and II molecules, presented on bioengineered Th1-polarized donor cells (AlloStim), and the recipient's T-cell and NK cell receptors at the immune synapse. This interaction is the fundamental mechanism of the AlloStim immunotherapy, designed to trigger a potent "mirror effect" where the host's immune system recognizes and rejects the foreign cells. By presenting intentionally mismatched MHC complexes, the therapy induces a controlled rejection response that releases high levels of Type 1 cytokines, such as interferon-gamma and TNF-alpha. These cytokines activate the patient's own innate and adaptive immune cells, including NK cells and T cells, which then infiltrate the tumor microenvironment. This process effectively converts "cold" tumors into "hot" tumors and promotes an in-situ vaccination effect against the patient's own tumor antigens. The target is primarily investigated for the treatment of chemotherapy-refractory metastatic solid tumors, such as colorectal and liver cancer, often in combination with checkpoint inhibitors like avelumab. This approach aims to overcome tumor-induced immunosuppression and elicit a durable, systemic anti-tumor immune response.
