Overview

Alpha-1,4-linked glucan polysaccharides are complex carbohydrates composed of glucose monomers linked by alpha-1,4-glycosidic bonds, serving as a primary energy storage form in plants as starch and animals as glycogen (PubChem, CID 439207). In the human body, these polysaccharides are broken down into glucose by enzymes such as alpha-amylase and alpha-glucosidase to provide energy for cellular processes (StatPearls, NBK557848). The regulation of this breakdown is a major therapeutic focus in the management of type 2 diabetes, where inhibitors like acarbose are used to slow digestion and mitigate postprandial blood sugar spikes. Additionally, the pathological accumulation of these glucans in lysosomes due to a deficiency in the enzyme acid alpha-glucosidase results in Pompe disease, a debilitating neuromuscular disorder (NIH, Pompe Disease). Treatment for such conditions involves enzyme replacement therapy to facilitate the degradation of the stored polysaccharides. These molecules also play a role in the structural integrity of bacterial biofilms, particularly in the oral cavity where they contribute to the development of dental caries (PubMed, PMID 23203237).
