The alpha-1D adrenergic receptor (α1D adrenoreceptor), encoded by the ADRA1D gene, is a G protein-coupled receptor primarily expressed in vascular smooth muscle, especially epicardial coronary arteries. It mediates vasoconstriction and regulates systemic blood pressure. Selective antagonism of α1D may offer coronary vasodilation without negative cardiac side effects.
Activation by catecholamines leads to Gq/11 protein activation, resulting in increased intracellular calcium influx and smooth muscle contraction, particularly in vascular tissues. Antagonists block this receptor, inhibiting vasoconstriction.
