Alpha-6 beta-2-containing neuronal nicotinic acetylcholine receptors (α6β2* nAChRs) are specialized pentameric ligand-gated ion channels primarily localized within the catecholaminergic pathways of the central nervous system, specifically the substantia nigra and ventral tegmental area [PMID: 15140937]. These receptors are unique because they are almost exclusively expressed on the terminals of dopaminergic neurons, where they exert powerful control over the release of dopamine in the striatum and nucleus accumbens [PMID: 17015497]. Due to this specific localization, α6β2* nAChRs are considered high-priority therapeutic targets for Parkinson's disease, as their modulation may help preserve dopaminergic function or alleviate motor symptoms [PMID: 21490162]. Pharmacological intervention typically involves the use of small molecule agonists or partial agonists, though achieving selectivity over the more ubiquitous α4β2 subtype remains a significant medicinal chemistry challenge [PMID: 11834714]. Research continues to explore selective peptide toxins and novel synthetic ligands to better understand the therapeutic potential of this receptor in neurodegenerative and psychiatric conditions [PMID: 22040696].
