Alpha-actinin-1 (ACTN1) is a non-muscle cytoskeletal protein belonging to the spectrin superfamily, primarily functioning as a **F-actin cross-linking protein** that anchors actin filaments to various intracellular structures[1][2][3][4]. Expressed in non-muscle cells, ACTN1 is localized along microfilament bundles and adherens-type junctions, where it tethers actin to the plasma membrane and stabilizes cell–cell adhesion. It plays central roles in **cytoskeletal organization, cell adhesion, migration, and signal transduction**. In immune cells such as T lymphocytes, it mediates cytoskeletal interactions for immune synapse formation. Disease associations include bleeding disorders (notably macrothrombocytopenia)[3], and tumor-specific splicing of ACTN1 has been observed in certain cancers, suggesting biomarker utility[1].
