Overview

Alpha-adducin is a cytoskeletal protein encoded by the ADD1 gene that is ubiquitously expressed and functions as a key assembly and regulatory factor at the spectrin-actin membrane skeleton in multiple cell types, especially red blood cells and neurons. It forms heterodimers or heterotetramers with beta- or gamma-adducin subunits and regulates actin filament capping, spectrin recruitment, and membrane stabilization, as well as participating in cell signaling through phosphorylation and calmodulin binding. Alpha-adducin variants, particularly the Gly460Trp polymorphism, are implicated in the pathogenesis of essential hypertension via sodium homeostasis effects in renal tubules. In addition, alpha-adducin can play roles in cell motility, division, and intracellular signaling, with emerging data supporting additional functions in tumorigenesis and metastasis via cytoskeletal reorganization. There are currently no direct pharmacological inhibitors or activators of alpha-adducin in clinical use, and it is not considered a traditional therapeutic target such as a receptor or enzyme, but its variants are clinically relevant as biomarkers for hypertension and potentially for certain cancers.
