Overview

The alpha-beta tubulin heterodimer is the fundamental structural subunit of microtubules, which are dynamic cytoskeletal filaments essential for various cellular processes (UniProt, 2024). Each heterodimer consists of one alpha-tubulin and one beta-tubulin polypeptide, both of which bind GTP to regulate polymerization and stability (PubMed, 2022). During mitosis, microtubules form the mitotic spindle, which is responsible for the accurate segregation of chromosomes into daughter cells (StatPearls, 2023). Because of its critical role in cell division, the tubulin heterodimer is a major therapeutic target in oncology (NIH, 2023). Drugs targeting tubulin, such as taxanes and vinca alkaloids, interfere with microtubule dynamics, leading to cell cycle arrest at the G2/M phase and subsequent apoptosis (PubChem, 2024). Beyond cancer, tubulin is also a target for treating inflammatory conditions like gout through the inhibition of leukocyte migration (StatPearls, 2023). However, since microtubules are also vital for axonal transport in neurons, tubulin-binding agents often cause dose-limiting peripheral neuropathy (PubMed, 2021).
