Alpha-centractin (ACTR1A) is a highly conserved actin-related protein that serves as the major structural subunit of the dynactin complex, a multi-protein assembly crucial for intracellular transport by linking cytoplasmic dynein to its cargo[1][2][3][5]. ACTR1A is present as 8–13 copies per dynactin complex and forms a filamentous core, sharing about 60% amino acid identity with conventional actin[1][2][5]. The dynactin complex, through ACTR1A, is vital for multiple cellular processes such as ER-to-Golgi transport, lysosome and endosome movement, mitotic spindle formation, chromosome movement, nuclear positioning, and axonogenesis[1][3][5]. Apart from its canonical cytoskeletal and transport functions, ACTR1A has been implicated in immune regulation (notably as a TLR2 signal cascade regulator) and may play a role in tumorigenesis as genetic alterations have been observed in certain cancers[3]. No drugs currently target ACTR1A directly, and it is not generally considered a therapeutic target[1][3][5].
