The alpha-fetoprotein promoter is a regulatory DNA sequence upstream of the alpha-fetoprotein (AFP) gene, which controls AFP transcription primarily in fetal liver and yolk sac, and is tightly repressed postnatally in most tissues. It contains tissue-specific response elements and is activated in certain pathological conditions, especially liver cancers, allowing it to be harnessed in biotechnology and gene therapy to drive selective expression of transgenes in hepatocytes or AFP-expressing tumor cells. This approach enables the study of gene function in specific tissues or the targeting of therapeutic genes to tumor cells with minimal activity in normal adult tissues.
