Alpha-glucosidase I (GluI) is a key enzyme in the eukaryotic N-glycosylation processing pathway, playing a critical role in glycoprotein processing within the endoplasmic reticulum. This enzyme catalyzes the first trimming step of newly synthesized glycoproteins, which is essential for proper protein folding and quality control. It selectively hydrolyzes terminal non-reducing (1→4)-linked α-glucose residues to release a single α-glucose molecule from the unique glycan substrate Glc3Man9GlcNAc2. Inhibition of GluI activity has been shown to impact the infectivity of enveloped viruses, suggesting potential therapeutic applications in antiviral treatments.
