Alpha-protein kinase 1 (ALPK1) is a serine/threonine protein kinase that plays a critical role in innate immunity by detecting bacterial pathogen-associated molecular pattern metabolites (PAMPs), particularly ADP-D-glycero-beta-D-manno-heptose (ADP-Heptose), and initiating downstream inflammatory signaling pathways. It is also involved in vesicular trafficking, protein sorting, and has been implicated in several diseases including cancer, gout, chronic kidney disease, myocardial infarction, and type 2 diabetes mellitus.
ALPK1 phosphorylates TIFA, activating the NF-kappa-B pathway.
