Alpha-secretases are a family of proteolytic enzymes, primarily ADAM10, that cleave amyloid precursor protein (APP) within its transmembrane region. This cleavage occurs within the sequence that would otherwise generate amyloid beta (Aβ) peptide. By cleaving APP at this site, alpha-secretase precludes Aβ formation and thus participates in the non-amyloidogenic pathway of APP processing, making it a therapeutic target for Alzheimer's disease.
Enhancement of alpha-secretase activity promotes non-amyloidogenic processing of APP, reducing amyloid-beta production.
