Overview

The Alpha-synuclein gene promoter is the regulatory DNA sequence that controls the expression of the SNCA gene, which encodes the alpha-synuclein protein. This promoter is a primary therapeutic target because the overproduction of alpha-synuclein is a central driver in the pathogenesis of Parkinson's disease and other synucleinopathies (Mittal et al., 2017) [1]. Genetic variations within the promoter, such as the Rep1 microsatellite, have been clinically linked to an increased risk and earlier onset of neurodegenerative disorders (Pihlstrøm et al., 2018) [3]. Pharmacological modulation of this target often involves the use of beta-2 adrenergic receptor agonists, which have been shown to reduce SNCA transcription by altering histone acetylation patterns (specifically H3K27ac) at the promoter site (Mittal et al., 2017) [1]. Additionally, epigenetic strategies focusing on increasing DNA methylation at the SNCA promoter are being explored to silence gene expression in affected neurons (Desplats et al., 2011) [2]. Reducing alpha-synuclein levels at the source—transcription—is considered a promising approach to prevent the toxic aggregation of the protein into Lewy bodies. However, therapeutic interventions must carefully balance the reduction of toxic protein levels with the preservation of alpha-synuclein's physiological role in synaptic vesicle trafficking and neurotransmitter release (Theisler et al., 2021) [4].
