Overview

The alpha-synuclein (SNCA) gene promoter is a regulatory DNA sequence that controls the transcription of the SNCA gene, which encodes the alpha-synuclein protein. In neurodegenerative disorders such as Parkinson's disease (PD), the overexpression of alpha-synuclein leads to the formation of toxic aggregates known as Lewy bodies (Mittal et al., 2017). Genetic variations within the promoter, such as the Rep1 microsatellite, are known to increase PD risk by enhancing SNCA expression (Maraganore et al., 2006). Therapeutic strategies targeting this region aim to reduce the production of alpha-synuclein at the transcriptional level. For instance, beta-2 adrenoreceptor agonists like salbutamol have been shown to reduce SNCA transcription by modulating histone acetylation at the promoter (Mittal et al., 2017). Furthermore, epigenetic modifications, specifically DNA methylation within the promoter and the closely associated Intron 1, serve as critical regulators of gene dosage and potential targets for intervention (Jowaed et al., 2010).

Modulation of SNCA gene transcription through the recruitment of transcription factors or alteration of epigenetic marks, such as histone acetylation or DNA methylation, to reduce alpha-synuclein protein production (Mittal et al., 2017; Jowaed et al., 2010).
