Overview

Alpha-synuclein-induced neuroinflammation refers to the inflammatory response in the central nervous system (CNS) triggered by the accumulation and aggregation of alpha-synuclein, a presynaptic neuronal protein. This process is central to the pathogenesis of Parkinson’s disease (PD) and other synucleinopathies. Alpha-synuclein aggregates are released from neurons into the extracellular space, where they interact with microglia, initiating a pro-inflammatory signaling cascade involving TLR2/TLR4, NF-κB, and the NLRP3 inflammasome, leading to the release of pro-inflammatory cytokines and subsequent neuronal death.
