Overview

Aminoacyl-tRNA accommodation at the A site of the ribosome is a critical step in protein synthesis during translation. It refers to the process by which an aminoacyl-tRNA (aa-tRNA)—a tRNA molecule charged with its corresponding amino acid—moves from a partially bound, pre-accommodation state (A/T state) into a fully accommodated position (A/A state) within the ribosomal A site. This movement positions the 3′-CCA end of aa-tRNA into the peptidyl transferase center (PTC), enabling peptide bond formation with the growing polypeptide chain. The process is multistep and involves significant conformational changes in both tRNA and ribosomal RNA. Proper accommodation ensures fidelity during translation: only correctly paired aa-tRNAs are efficiently accommodated for peptide bond formation.
