Overview

Aminosteroidal nondepolarizing neuromuscular blocking agents are drugs used primarily in anesthesia to induce skeletal muscle relaxation by competitively and reversibly antagonizing the muscle-type nicotinic acetylcholine receptor at the neuromuscular junction. They prevent acetylcholine from binding, blocking neuromuscular transmission and causing flaccid paralysis in skeletal muscle. Examples include rocuronium, vecuronium, and pancuronium. These agents are essential for endotracheal intubation and surgical muscle relaxation and are associated with unique risks such as anaphylaxis and the need for supportive measures (e.g., ventilatory support) during use

Mechanism of action

Competitive antagonism of the muscle-type nicotinic acetylcholine receptor (nAChR) at the neuromuscular junction, preventing acetylcholine from activating the receptor and thereby causing skeletal muscle relaxation
