Overview

AMP-activated protein kinase beta 1 subunit (AMPK β1, gene symbol PRKAB1) is one of two known β regulatory subunits of the AMPK enzyme complex, responsible for scaffolding the complex together and mediating its interactions with cellular structures. AMPK is a highly conserved serine/threonine kinase, integral to cellular energy sensing. The β1 subunit contains a carbohydrate-binding module and a conserved C-terminal domain that tethers the alpha and gamma subunits. The AMPK complex (α, β, and γ subunits) responds to increased cellular AMP/ATP ratios by activating pathways to enhance energy production and reduce energy consumption. Targeting AMPK, and by extension its β1 subunit, has therapeutic potential in metabolic syndrome, type II diabetes, obesity, cancer, and cardiovascular disease, with metformin and thiazolidinediones known to exert some effects through this pathway[1][6][7][8].

Mechanism of action

Allosteric activation of the AMPK complex by AMP or small molecule mimetics Direct activation of AMPK by phosphorylation at its alpha subunit (drugs and endogenous kinases activate the complex containing the β1 subunit) Indirect activation by metabolic stress (AMP/ATP ratio alteration)
