Overview

Amphiphysin is a membrane-associated adaptor protein characterized by the presence of a BAR (Bin/Amphiphysin/Rvs) domain, which enables the detection and induction of membrane curvature essential for intracellular trafficking events. Amphiphysin 1 (AMPH1) is primarily expressed in neuronal tissues, where it functions as a key regulator of clathrin-mediated endocytosis at synapses by recruiting and activating dynamin, thereby driving synaptic vesicle fission and recycling[1][2][3]. It contains additional binding sites for clathrin and adaptor protein 2 (AP-2), and its interactions are tightly regulated by phosphorylation/dephosphorylation. In the context of human disease, autoantibodies directed against amphiphysin are a hallmark of paraneoplastic neurological syndromes such as stiff-person syndrome associated with breast cancer[3]. Although amphiphysin plays a critical cellular role, there are currently no therapeutic drugs targeting this protein directly, and it is not considered a canonical therapeutic target like a receptor or enzyme.
