Overview

The Amylin 3 receptor (AMY3) is a heterodimeric G protein-coupled receptor formed by the association of the calcitonin receptor (CTR) and the receptor activity-modifying protein 3 (RAMP3) (IUPHAR/BPS Guide to Pharmacology, 2023). It serves as a primary mediator for the physiological effects of amylin, a peptide hormone co-secreted with insulin by pancreatic beta cells to regulate glucose metabolism (PubMed: 31433984). Activation of AMY3, particularly in the area postrema of the hindbrain, plays a critical role in regulating energy homeostasis by inducing satiety and slowing gastric emptying (StatPearls, 2023). In addition to appetite control, AMY3 signaling suppresses postprandial glucagon secretion, which prevents excessive hepatic glucose production (UniProt: P30988). Because of its central role in metabolic regulation, AMY3 is a significant therapeutic target for the treatment of obesity and type 2 diabetes. Pharmaceutical agents such as pramlintide and the long-acting analog cagrilintide act as agonists at this receptor complex to promote weight loss and improve glycemic control (PubMed: 33901487). Beyond metabolism, the receptor is also involved in bone remodeling, where it may influence osteoclast activity (PubMed: 22431003).
