The amylin receptor is a heterodimeric receptor complex composed of the calcitonin receptor (CTR) and one of three receptor activity-modifying proteins (RAMPs): RAMP1, RAMP2, or RAMP3. These combinations form distinct receptor subtypes AMY1, AMY2, and AMY3, each with varying affinities for amylin and other ligands. The amylin receptor is widely distributed in the central nervous system and peripheral tissues, playing crucial roles in regulating appetite, gastric emptying, glucose metabolism, and body fluid balance. It is implicated in the pathophysiology of type II diabetes mellitus, obesity, and Alzheimer's disease, making it a potential therapeutic target.
Activation of the amylin receptor leads to downstream signaling cascades that mediate effects on gastric emptying, appetite, and glucose metabolism.
