Overview
Pyroglutamate-modified amyloid beta (Aβ) refers to N-terminally truncated forms of the amyloid beta peptide in which the third amino acid residue (glutamate) is cyclized to form pyroglutamate. These modified peptides exhibit increased cytotoxicity and aggregation propensity compared to unmodified Aβ, making them a key therapeutic target for Alzheimer's disease. They are formed via enzymatic modification of truncated Aβ peptides by glutaminyl cyclase or cathepsin B.
