Amyloid beta A4 precursor protein-binding family A member 3 (APBA3) is an adapter protein of the X11/Mint family that interacts with the cytoplasmic domain of the Alzheimer’s disease amyloid precursor protein (APP) and plays important roles in signal transduction, cellular metabolism, and inflammation. In macrophages, APBA3 (also known as Mint3) binds and sequesters FIH-1, leading to enhanced HIF-1 activity under normoxic conditions, promoting glycolytic ATP production and supporting energy-intensive immune functions such as cytokine production and chemotaxis. APBA3 thereby acts as a nodal mediator linking metabolic regulation with immune responses and cell motility. It is implicated both in neurodegenerative conditions (Alzheimer’s disease) and cancer progression, and is regarded as a potential therapeutic target for modulating aberrant macrophage activity in diseases involving inflammation and metastasis.
