Amyloid beta (Aβ) oligomer formation refers to the process by which monomeric Aβ peptides aggregate into small, soluble assemblies known as oligomers. These oligomers are considered key neurotoxic species in the pathogenesis of Alzheimer’s disease and are distinct from larger, insoluble amyloid fibrils that form plaques. The aggregation of Aβ monomers into oligomers is an early event in amyloidogenesis and is closely linked to synaptic dysfunction, neuronal toxicity, and cognitive deficits observed in Alzheimer’s disease. Oligomeric forms are more neurotoxic than mature fibrils or plaques; they disrupt synaptic function by interacting with cell membranes or specific receptors at synapses, leading to altered calcium homeostasis and excitotoxicity.
