Overview

The pyroglutamated form of amyloid beta peptide refers to amyloid beta (Aβ) peptides that have undergone N-terminal truncation and subsequent modification, where the exposed glutamate residue at position 3 or 11 is cyclized into a pyroglutamate (pE) by the enzyme glutaminyl cyclase. This post-translational modification results in Aβ species such as AβpE3-42 and AβpE11-42, which are highly relevant in Alzheimer’s disease (AD) pathology due to their unique biochemical properties and pathological roles. These modified forms exhibit enhanced aggregation kinetics, increased stability, and higher cytotoxicity. Therapeutic targeting includes antibody therapies and inhibition of glutaminyl cyclase.
