Amyloid beta precursor protein binding family B member 3 (APBB3), also called Fe65L2, is an intracellular adaptor protein primarily localized to the cytoplasm and sometimes the nucleus[1][3][5][7]. It is a member of the APBB protein family, which is characterized by WW and phosphotyrosine‐binding (PTB) domains[1]. APBB3 binds to the intracellular domain of the amyloid precursor protein (APP) as well as APP-like proteins, influencing APP signaling, internalization, and processing—a central pathway implicated in Alzheimer's disease pathogenesis[1][3][5][7]. Alternative splicing of APBB3 produces multiple isoforms with varying binding and localization properties[1]. Additionally, APBB3 has candidate roles in synaptic development and neurodevelopmental processes. Although associated with disease pathways (notably neurodegeneration), it is not an established therapeutic target[1][3][5].
