Overview

AN1-type zinc finger protein 3 (ZFAND3) is a nuclear DNA-binding protein and transcription factor that contains both A20 and AN1 zinc finger domains[1][3]. ZFAND3 acts within a nuclear protein complex to activate the transcription of invasion-related genes, including those involved in cell adhesion and migration, making it a crucial driver of glioblastoma invasion and possibly other invasive cancers[1][3][4]. It is also physiologically expressed in reproductive tissues where it contributes to germ cell maturation[2]. In cancer biology, ZFAND3 overexpression enhances the motility and invasiveness of glioblastoma cells, while knockout or downregulation impairs tumor infiltration into neighboring tissue[1][3][4]. ZFAND3 regulates the promoters of several key genes such as COL6A2, FN1, and NRCAM and interacts with various nuclear proteins, emphasizing its role as a transcriptional regulator rather than a traditional therapeutic drug target[1][3][4]. There are currently no drugs known to directly target ZFAND3, and it is not considered a typical therapeutic target class like a receptor or enzyme[7][8].
