The anal mucosa is the innermost tissue layer lining the anal canal, serving as a critical physiological barrier between the internal environment and the external world. Understanding the unique histological and immunological properties of the anal mucosa is vital for developing effective localized drug delivery systems and diagnostic tools for colorectal health.
Drugs applied to the anal mucosa act through various molecular mechanisms depending on the active ingredient, such as blocking voltage-gated sodium channels to provide local anesthesia, activating glucocorticoid receptors to reduce inflammation, or donating nitric oxide to induce smooth muscle relaxation (StatPearls, 2023; PubMed, 2021).
