Overview

An analgesic agent is a pharmacological term used to describe any member of the group of drugs used to achieve analgesia, or relief from pain (NIH NCI, 2024). It is not a single molecular target or receptor but rather a functional classification of medications that target a wide array of biological systems, including the opioid system, the arachidonic acid pathway, and various ion channels (Wikipedia, 2023). These agents are critical for the management of pain associated with trauma, surgery, and chronic conditions like cancer or arthritis. Because this term refers to a therapeutic class rather than a specific protein, enzyme, or receptor, it is considered an incorrect or overly broad entry for a specific drug target. The therapeutic application of analgesics is often complicated by severe side effects such as respiratory depression for opioids or gastrointestinal damage for non-steroidal anti-inflammatory drugs (StatPearls, 2023).

Mechanism of action

Analgesic agents act through various mechanisms including the inhibition of cyclooxygenase enzymes to reduce prostaglandin synthesis, the activation of mu-opioid receptors in the central nervous system, and the modulation of voltage-gated ion channels to inhibit pain signaling (StatPearls, 2023).
