Anaphase-promoting complex subunit 11 (ANAPC11) is a core catalytic component of the anaphase-promoting complex/cyclosome (APC/C), a large multi-subunit E3 ubiquitin ligase essential for cell cycle control. ANAPC11 contains a RING-H2-finger domain responsible for mediating the transfer of ubiquitin from E2 conjugating enzymes to substrate proteins, targeting them for proteasomal degradation. The APC/C complex, with ANAPC11 acting as a central ubiquitin ligase, governs the timely degradation of cell cycle regulators such as cyclins and securin, thereby driving progression through mitosis and the G1 phase. Dysregulation or overexpression of ANAPC11 has been implicated in cancer progression (e.g., urothelial bladder cancer and hepatocellular carcinoma), where it can promote cancer cell proliferation, invasion, and metastasis by increasing the ubiquitination and degradation of key regulators such as FOXO3. Due to its enzymatic role in cell cycle regulation and oncogenic activity in certain cancers, ANAPC11 is regarded as a candidate therapeutic target, although no clinically approved drugs currently act specifically on it.
Drugs or experimental agents targeting ANAPC11 would act by modulating ubiquitin ligase activity—generally by inhibiting the ubiquitination and proteasomal degradation of cell cycle-regulating proteins (e.g., cyclins, securin, FOXO3).
