Overview

Anaphase-promoting complex subunit 2 (ANAPC2) is a cullin-family protein forming the catalytic core of the anaphase-promoting complex/cyclosome (APC/C), which is a large multi-subunit E3 ubiquitin ligase that orchestrates the metaphase to anaphase transition and exit from mitosis by targeting specific cell cycle regulators—such as securin and mitotic cyclins—for ubiquitin-mediated proteolysis. This process ensures orderly chromosome segregation and cell cycle progression. ANAPC2 interacts with APC11 to form the minimal catalytic module required for ligase activity; dysregulation of APC/C and ANAPC2 is implicated in tumorigenesis and other proliferative diseases.

Mechanism of action

Inhibition of APC/C function to prevent cell cycle progression, leading to cell cycle arrest and apoptosis in cancer cells; Disruption of APC2:APC11 complex formation impairs ubiquitin ligase activity. Most inhibitors affect APC/C by preventing substrate recognition or coactivator interaction, indirectly modulating ANAPC2's catalytic role.
