Overview

Anaphase-promoting complex subunit 4 (ANAPC4) is a core component of the anaphase-promoting complex/cyclosome (APC/C), a multi-subunit E3 ubiquitin ligase essential for cell cycle regulation. The APC/C complex orchestrates the transition from metaphase to anaphase by targeting key regulatory proteins such as mitotic cyclins and anaphase inhibitors for ubiquitination, leading to their degradation by the 26S proteasome[1][2][3][4]. ANAPC4 itself does not have direct catalytic activity but is crucial for the assembly and function of the APC/C and thereby for proper cell cycle progression. Mutations or altered expression of ANAPC4 are associated with cell cycle dysregulation, tumor progression, and therapeutic resistance, especially in various cancers. The gene is conserved across eukaryotes, and disruption of its function affects mitosis and can lead to developmental disorders such as Adams-Oliver syndrome[3].
