Anaphase-promoting complex subunit CDC26 (CDC26) is a protein that acts as an essential component of the Anaphase-promoting complex/cyclosome (APC/C), a large E3 ubiquitin ligase that controls cell cycle transitions by targeting key cell cycle regulators for ubiquitin-mediated proteolysis and subsequent degradation[3][4][5][7][10]. CDC26 itself has no apparent enzymatic activity, but instead is critical for the structural assembly and stabilization of the complex—especially its interaction with the TPR domain of APC6[1][6]. This function is conserved from yeast to humans, and loss of CDC26 disrupts APC/C activity leading to impaired cell division and growth defects. CDC26 is implicated in diseases associated with cell cycle dysfunction, notably cancer[5]. As of now, no clinically approved drugs are known to directly target CDC26 or the APC/C for therapeutic intervention, although the complex itself is a focal point for cell cycle inhibitor development, making CDC26 part of a therapeutically relevant molecular family.
