Overview

Anaplastic Lymphoma Kinase (ALK) is a receptor tyrosine kinase involved in cell growth and differentiation. Aberrant ALK activity, often due to gene fusions or activating mutations, drives oncogenesis in several cancers, including lymphoma, lung cancer, and neuroblastoma. It functions as a "dependence receptor", where, without ligand, it induces caspase‐3 mediated apoptosis. Small-molecule inhibitors targeting ALK are used clinically for treating ALK-positive cancers.
