Overview

Anchoring proteins is a general term for a heterogeneous group of proteins that function to spatially and temporally organize other proteins, especially signaling enzymes, within specific subcellular compartments. The most well-studied family is the A-kinase anchoring proteins (AKAPs), which tether protein kinase A (PKA) and other signaling molecules to defined locations, facilitating precisely localized signaling events[2][3][4][6][7][10][12]. Anchoring proteins are crucial for the functional compartmentalization of signal transduction pathways, permitting diverse and specific cellular responses to stimuli[2][3][4][6][7][10][12]. The class encompasses proteins anchored by lipid moieties (lipid-anchored proteins, such as GPI-anchored and fatty acid–anchored proteins), which can also mediate signaling, adhesion, and cell structure roles[1][5][13]. The target “anchoring proteins” is imprecise, does not refer to a singular molecular entity, and is not directly considered a therapeutic target as a class[2][3][6][7][10][12]. Specific members of this class, such as individual AKAPs, may be explored for targeted intervention in the future.
