Overview

Andes virus glycoprotein Gn is a critical structural component of the Andes virus (ANDV) envelope, forming a heterodimeric spike complex with glycoprotein Gc [1, 8]. It plays a primary role in viral attachment to host cell receptors, such as beta-3 integrins (ITGAV/ITGB3) and protocadherin-1, facilitating the initial stages of viral entry [3, 5, 13]. Beyond its structural role, the cytoplasmic tail of Gn is involved in modulating the host's innate immune response by inhibiting interferon signaling and inducing mitophagy to degrade MAVS [3]. As a major surface antigen, Gn is a key target for the development of neutralizing monoclonal antibodies and vaccines aimed at preventing Hantavirus Cardiopulmonary Syndrome (HCPS), a severe respiratory disease with high mortality rates [2, 11]. Experimental therapies focusing on Gn, such as the DNA vaccine pWRG/AND-M and various monoclonal antibodies, aim to block viral entry or assembly [2, 5, 12]. Despite its importance as a therapeutic target, no specific treatments or vaccines targeting Gn are currently approved for clinical use [4, 11].
