Overview

Androgen-dependent TFPI-regulating protein (ADTRP) is a membrane-associated enzyme encoded by the ADTRP gene (previously C6orf105) and is primarily expressed in endothelial cells. It is inducible by androgens and plays a critical role in regulating the expression and activity of tissue factor pathway inhibitor (TFPI), the major inhibitor of the tissue factor-dependent coagulation pathway. ADTRP also demonstrates hydrolase activity specific for fatty acid esters of hydroxy fatty acids (FAHFAs), contributing to anti-diabetic and anti-inflammatory lipid metabolism. Functionally, ADTRP supports vascular development and barrier integrity, in part by modulating Wnt signaling and matrix metalloproteinase-9 (MMP-9) expression. Genetic variants of ADTRP are associated with several vascular diseases, including coronary artery disease and thrombosis, and its deficiency can cause abnormal angiogenesis, vascular leakage, and increased inflammation.
