Overview

Androgen receptor regulated long noncoding RNA 1 (ARLNC1) is a prostate-specific long noncoding RNA directly regulated by androgen receptor (AR) signaling. ARLNC1 forms a positive feedback loop by stabilizing AR transcript via RNA-RNA interaction, which potentiates AR signaling and enhances prostate cancer cell proliferation and survival[1][2][4][5][6][7][8]. Knockdown of ARLNC1—using RNA interference or antisense oligonucleotides—suppresses AR levels, global AR signaling, and tumor growth both in vitro and in vivo[1][6]. ARLNC1 is under investigation as a therapeutic target and as a biomarker for AR-driven prostate cancer[1][6]. No approved drugs currently target ARLNC1 directly, though RNA-targeted approaches are in development.

Mechanism of action

Antisense oligonucleotides (ASOs): inhibit ARLNC1, leading to reduced AR transcript stability and signaling, suppressed prostate cancer growth

03

Biological functions

Regulation of androgen receptor signalingRegulation of cell proliferationRegulation of apoptosisRegulation of DNA damage response
04

Disease associations

Cancer (especially prostate cancer and castration-resistant prostate cancer)
05

Safety considerations

Possible tissue-specific effects, as ARLNC1 is expressed almost exclusively in prostate tissueNeed for further characterization of off-target effects when using nucleotide-based therapies
06

Interacting drugs

Antisense oligonucleotides (ASOs)
07

Biomarkers

ARLNC1 expression in prostate tissue as a mechanistic/predictive biomarker (potential utility for advanced prostate cancer and as a marker for resistance to therapies targeting AR signaling)
