An angiogenesis inhibitor is a substance that blocks the formation of new blood vessels (angiogenesis). These inhibitors can be endogenous (naturally occurring in the body) or exogenous (administered as drugs or derived from diet). The primary therapeutic use of angiogenesis inhibitors is to restrict the blood supply to tumors, thereby inhibiting their growth and metastasis. Unlike traditional cancer therapies that target tumor cells directly, angiogenesis inhibitors act on the endothelial cells lining blood vessels, preventing them from proliferating and forming new vasculature needed for tumor survival.
Angiogenesis inhibitors work by interfering with various steps in the formation of new blood vessels, primarily by blocking pro-angiogenic factors such as VEGF, FGF, PDGF, and TGF-β. This can involve direct binding of antibodies to these factors, inhibition of receptor tyrosine kinase activity, disruption of integrin-mediated adhesion, or modulation of immune responses to reduce pro-angiogenic cytokine production.
