Angiomotin-like protein 2 (AMOTL2) is a member of the motin family of proteins, which are involved in the regulation of endothelial cell morphology, motility, and proliferation[1][3]. AMOTL2 functions as a scaffold and adaptor protein at cell–cell junctions, especially tight junctions, and is a crucial activator of the Hippo signaling pathway through direct binding and activation of the LATS2 kinase[2][3]. In this role, AMOTL2 promotes phosphorylation and cytoplasmic sequestration of the transcriptional coactivators YAP and TAZ, thereby inhibiting their ability to drive pro-proliferative gene expression. AMOTL2 also participates in actin cytoskeleton organization, maintaining cell architecture and epithelial sheet integrity[3]. It is implicated in processes such as angiogenesis, tissue growth, and cancer progression, largely through its control of Hippo pathway signaling and regulation of contact inhibition[1][2][3]. Functions as a scaffold for Hippo pathway proteins, promoting activation of LATS2 kinase and suppression of YAP/TAZ transcriptional coactivator activity. Inhibits Wnt/β-catenin pathway by sequestering CTNNB1 (β-catenin). Sequesters YAP/TAZ in the cytoplasm at tight junctions to repress their activity.
