Overview

Tie2 is a receptor tyrosine kinase (RTK) that is part of the angiopoietin–Tie signaling system, which is vascular-specific and essential for normal vascular development. Expressed primarily on endothelial cells lining blood vessels and lymphatics, Tie2 activation by its ligands triggers downstream signaling pathways such as PI3K/AKT that promote cell survival, quiescence, barrier function, and anti-inflammatory effects. Dysregulation of the angiopoietin-Tie pathway contributes to diseases such as cancer (tumor angiogenesis), inflammation-related vascular leakage syndromes, diabetic retinopathy, sepsis-induced endothelial dysfunction.
