Overview

Angiopoietin-4 (ANGPT4) is a secreted glycoprotein and member of the angiopoietin family, encoded by the human ANGPT4 gene on chromosome 20p13[1][4]. It shares structural homology with ANGPT1 and ANGPT2, including a coiled-coil domain and a fibrinogen-like domain, permitting binding to the endothelial-specific TIE2 receptor[1]. ANGPT4 regulates angiogenesis, vascular maturation, and lymphatic vessel growth. Its expression is upregulated under hypoxic conditions and in various tumors, where it contributes to cancer cell proliferation, migration, metastasis, and the modulation of tumor microenvironment[1][4]. ANGPT4-mediated signalling involves pathways such as PI3K-Akt, MAPK, ERK, Ras, and Hedgehog, directly or via TIE2 receptor activation[1]. Recent studies highlight its potential roles in cardiac repair and possible metabolic regulation through effects on endothelial cell function[1]. ANGPT4 is a promising therapeutic and biomarker target, but poses safety risks related to vascular and lymphatic system homeostasis when inhibited[1].

Mechanism of action

Targeting ANGPT4 would likely modulate angiogenesis by interfering with its interaction with the TIE2 receptor and/or downstream signalling pathways such as PI3K-Akt, ERK, MAPK, Ras, and Hedgehog signaling.
