Overview

Angiopoietin-like protein 1 (ANGPTL1) is a secreted glycoprotein and the founding member of the angiopoietin-like protein family. Unlike classical angiopoietins, ANGPTL1 does not bind the Tie1 or Tie2 receptors and is considered to act through distinct mechanisms and possibly alternative receptors such as leukocyte immunoglobulin-like receptor B2 (LILRB2). ANGPTL1 is a potent inhibitor of angiogenesis and tumor metastasis, partly by blocking endothelial cell migration and proliferation and by exerting anti-apoptotic effects via ERK1/2 and Akt-1 signaling. It has been found to be downregulated in multiple human cancers, and reduced expression is associated with increased metastasis and poorer clinical prognosis. In addition to its key anti-angiogenic effects, ANGPTL1 plays roles in inflammation, lipid metabolism, and hematopoiesis, and remains under investigation for its potential as a biomarker and therapeutic tool in cancer and vascular pathologies
